    Ingested insecticide to control Aedes aegypti: developing a novel dried attractive toxic sugar bait device for intra-domiciliary control

Background: Illnesses transmitted by Aedes aegypti (Linnaeus, 1762) such as dengue, chikungunya and Zika comprise a considerable global burden; mosquito control is the primary public health tool to reduce disease transmission. Current interventions are inadequate and insecticide resistance threatens the effectiveness of these options. Dried attractive bait stations (DABS) are a novel mechanism to deliver insecticide to Ae. aegypti. The DABS are a high-contrast 28 inch2 surface coated with dried sugar-boric acid solution. Aedes aegypti are attracted to DABS by visual cues only, and the dried sugar solution elicits an ingestion response from Ae. aegypti landing on the surface. The study presents the development of the DABS and tests of their impact on Ae. aegypti mortality in the laboratory and a series of semi-field trials. Methods: We conducted multiple series of laboratory and semi-field trials to assess the survivability of Ae. aegypti mosquitoes exposed to the DABS. In the laboratory experiments, we assessed the lethality, the killing mechanism, and the shelf life of the device through controlled experiments. In the semi-field trials, we released laboratory-reared female Ae. aegypti into experimental houses typical of peri-urban tropical communities in South America in three trial series with six replicates each. Laboratory experiments were conducted in Quito, Ecuador, and semi-field experiments were conducted in Machala, Ecuador, an area with abundant wild populations of Ae. aegypti and endemic arboviral transmission. Results: In the laboratory, complete lethality was observed after 48 hours regardless of physiological status of the mosquito. The killing mechanism was determined to be through ingestion, as the boric acid disrupted the gut of the mosquito. In experimental houses, total mosquito mortality was greater in the treatment house for all series of experiments (P \u3c 0.0001). Conclusions: The DABS devices were effective at killing female Ae. aegypti under a variety of laboratory and semi-field conditions. DABS are a promising intervention for interdomiciliary control of Ae. aegypti and arboviral disease prevention.[Figure not available: see fulltext.

    Percepción de la demanda y necesidades de formación en salud mental de los médicos de atención primaria

    ResumenObjetivoConocer la opinión de los médicos de atención primaria (AP) sobre la demanda asistencial en salud mental (SM), sus necesidades de formación y sus actitudes ante dichos trastornos.DiseñoEstudio observacional transversal.EmplazamientoAtención primaria.ParticipantesLos sujetos de estudio fueron médicos de AP. De un total de 1.191 médicos de AP en Castilla-La Mancha se seleccionó a 301 mediante muestreo aleatorio estratificado.Mediciones principalesSe elaboró un cuestionario autorrellenable y se indentificaron las siguientes variables: características profesionales y sociodemográficas, demanda percibida de problemas de SM y dificultades percibidas en su resolución, formación en SM, conocimientos sobre recursos especializados y actitudes hacia dichos trastornos.ResultadosLa mayoría (63,5%, intervalo de confianza [IC] del 95%, 57,6-69,4) consideró que los problemas mentales están relacionados con, al menos, el 20% de las consultas. Los problemas señalados como motivos de consulta muy frecuentes fueron: trastornos de ansiedad (75,9%), del estado de ánimo (73,5%) y somatomorfos (40,5%). La proporción que consideró suficiente el nivel de información sobre recursos especializados ascendió hasta el 63,6% (IC del 95%, 57,8-69,6). En cuanto a las actitudes, el 43,7% (IC del 95%, 37,7-49,8) opinó que los equipos de AP no pueden hacerse cargo de la demanda existente. Dicha opinión fue significativamente más frecuente en los médicos con más pacientes asignados, mayor presión asistencial, sin formación MIR, con menor demanda percibida, sin formación en SM y peor conocimiento de los recursos especializados.ConclusionesEn opinión de la mayoría de los médicos, los problemas mentales están relacionados con, al menos, una de cada 5 consultas. Una elevada proporción considera que los equipos de AP no pueden hacerse cargo de la demanda existente, sobre todo cuando la presión asistencia les elevada. La mayoría opina que debería haber mayor formación específica en SM y mayor coordinación con los servicios especializados.AbstractObjectiveTo know the opinion of the primary health care (PHC) physicians on the assistance demand in mental health (MH), their training necessities and their attitudes about this disorders.DesignObservational and transversal study.SettingPrimary care.ParticipantsThe subjects of the study were PHC physicians. From a total of 1191 PHC physicians in Castilla-La Mancha region, 301 were selected by random stratified sampling.Main measurementsA self-complimented questionnaire was elaborated, being identified the following variables: professional and sociodemographics characteristics, their perceived demands of MH problems, perceived difficulties in resolution, training in MH, knowledge about specialized resources and their attitudes about mental disorders.Results63.5% (95% confidence interval [CI], 57.6-69.4) considered that the mental problems are related with, at least, 20% of the consultations. About attitudes, 43.7% (95% CI; 37.7-49.8) said the PHC physicians cannot be taken charge of the existent demand. This opinion as significantly more frequents in physicians with more patient assigned, more assistance pressure, without postgraduate assistance formation (Spanish MIR system), with less perceived demand, without training in MH and worse knowledge of the specialized resources.ConclusionsIn opinion of most of the physicians the mental disorders are related with, at least, 1 of each 5 consultations. A high proportion considers that the teams of PHC cannot be taken charge of the existent demand, mainly when it is high the assistance pressure.Most says that more specific training should exist in MH and more coordination with the specialized services

    Kinetic and functional properties of human mitochondrial phosphoenolpyruvate carboxykinase

    The cytosolic form of phosphoenolpyruvate carboxykinase (PCK1) plays a regulatory role in gluconeogenesis and glyceroneogenesis. The role of the mitochondrial isoform (PCK2) remains unclear. We report the partial purification and kinetic and functional characterization of human PCK2. Kinetic properties of the enzyme are very similar to those of the cytosolic enzyme. PCK2 has an absolute requirement for Mn2+ ions for activity; Mg2+ ions reduce the Km for Mn2+ by about 60 fold. Its specificity constant is 100 fold larger for oxaloacetate than for phosphoenolpyruvate suggesting that oxaloacetate phosphorylation is the favored reaction in vivo. The enzyme possesses weak pyruvate kinase-like activity (kcat=2.7 s-1). When overexpressed in HEK293T cells it enhances strongly glucose and lipid production showing that it can play, as the cytosolic isoenzyme, an active role in glyceroneogenesis and gluconeogenesis

    Evaluation of Wedged Arterial Injection as a New Technique for Delivery of Experimental Therapeutic Sustances into the Porcine Pancreas

    Objectives. To prospectively evaluate the technical feasibility and efficacy of wedged arterial injection (WAI) as a potential route for experimental selective therapy to the pancreas of healthy pigs. Materials and Methods. Selective angiographies were completed in ten pigs under general anaesthesia. By superselective angiography, the catheter was inserted and wedged into the major pancreatic artery, blocking the blood flow. In order to evaluate the efficacy of the WAI method, a DNA-specific fluorescent dye (Hoechst 33258) was used. Results. Histological study revealed a uniform distribution of the fluorescent dye within the nuclei of the endocrine and exocrine pancreatic cells. Pancreatic and liver enzymes as well as histopathology of the pancreas were normal. Conclusion. WAI is a highly effective minimally invasive methodology to target the porcine pancreas. The findings suggest that WAI may contribute to developing preclinical assays of pancreas gene or cell-transfer therapies in swine model

    Analysis of the Calls Received during the COVID-19 Lockdown by the Mental Health Crisis Helpline Operated by the Professional College of Psychology of Aragon

    COVID-19 has had a direct impact on the physical and mental health of millions of people worldwide. Therefore, a Mental Health Crisis Helpline (MHCH) was set up and offered free of charge by the Professional College of Psychology of Aragon (COPPA) during the lockdown period. This research aims to study the reasons for the calls, to describe the population segments that used it, and to analyse the possible relationships between the variables studied. A total of 1411 calls were answered and 598 were registered. The main reasons for the calls were: anxiety symptoms, concern for a relative, previous mental health problems, conflicts, and depressive symptoms. Significantly more men called for anxiety symptoms (60.8% vs. 49.5%) versus more women calling regarding a chronic physical illness (3.5% vs. 0.7%), concern about a relative (22.7% vs. 12.4%), care guidelines (6% vs. 1.3%), and bereavement (6.2% vs. 2%). Calls regarding conflict increased slightly as the lockdown period progressed (p < 0.001; r = 0.15), in contrast with calls regarding previous psychological conditions and anxiety symptoms (p = 0.035; r = -0.09; p = 0.005; r = -0.12). These results highlight the intensive use of the MHCH, confirming the need for the implementation of specific psychological care resources in times of crisis. © 2022 by the authors.     Acute Effects of Muscular Fatigue on Vertical Jump Performance in Acrobatic Gymnasts, Evaluated by Instrumented Insoles: A Pilot Study

    The study of fatigue during training is becoming a very useful tool to avoid possible injuries not only during the training sessions but also during recovery time. Many researches have proved that concepts such as muscular fatigue and postactivation potentiation have a close relationship. With this aim, vertical jump can provide a very important information that can help to analyze the muscular fatigue that happened during this type of activity, mainly if the monitoring system is able to measure jumping parameters during their regular training session in their natural training environment. This study was performed with instrumented insoles called ECnsole. These insoles were tested with a group of twelve volunteers. In a tumbling surface, the participants performed a jumping protocol in three conditions: rest, fatigue-induced, and recovery. Using these validated insoles, the acrobatic gymnasts showed an inability to use the stretch-shortening cycle for improving vertical jumping performance after fatigue condition, although no deterioration of jump performance was found.Junta de Andalucía European Commission P10-TIC5997European Commissio

    Education knowledge transfer: Report on in-school placements needs and possibilities of the technologies

    The WP2 of the EKT project aimed at describing how in-school placement (ISP) is organised and developed in the initial teacher education in the countries participating in the EKT project (Austria, England, Ireland, Portugal and Spain). In order to achieve this goal, two main pr ocedures were developed: 1. The characterization of national ISP systems, based on the analysis of institutional documentation (legislation, tools, guides, regulations, protocols, reports...) identified and described by the EKT academic teams in each country 2. The identification of the perspectives and conceptions of academic and school mentors involved in ISP, working either Higher Education Institutions (HEI) where Initial Teacher Education (ITE) is developed or at the schools where ISP takes place. For this sake, a questionnaire was designed and applied to a sample of 347 mentors from the different countries involved in the project. The analysis of the different ITE systems enabled the identification of many common features. In fact, all the countries have ITE systems that involve the HEI and nursery/primary/ secondary schools where ISP takes place. In most situations, ITE involves consecutive programmes (at graduation and post-graduation levels). And despite some variation in the length of these consecutive programmes (6 + 3/4 semesters or 8 + 2/3 semesters), the extension of the whole ITE process is less variable, the same happening with the global number of ECTS units involved in any ITE process regardless of the country. In some countries (e.g., Spain), and concerning, mainly but not exclusively, nursery and primary school teachers’ preparation, ITE may take place at the graduation level. It is also worth mentioning the case of England, where two different teacher training paths can be found: HEI routes and School-led postgraduate (consecutive) routes that involve different kinds of programmes: • School-centred initial teacher training (SCITT) programmes (no salaried); • School Direct Training programmes (non-salaried); • School Direct Training programmes (salaried); • Teach First (salaried); • Postgraduate Teaching; Apprenticeship (PGTA) (salaried). No matter the country, ITE processes imply in-school placement. The characteristics of these placements may vary according not only to the features of each national educational system but also to the respective ITE programmes and the school level training refers to. In general ISP is developed under agreements established between HEI where ITE programmes are developed and schools where ISP takes place and involves both HEI teachers and school teachers as mentors. The selection of these school mentors varies from country to country as well as the role they play and their participation in the students´ evaluation. There also seems to be some variation regarding the tasks, the number of hours involved and the kind of materials students have to produce and deliver as a result of their practices during ISP. Despite this variation, it is possible to recognize that there is, in most cases, a concern with the promotion of a reflection on the practice by the use of adequate practices and instruments, such as the construction of portfolios that make possible a critical attitude towards the teaching activities developed. Executive summary The questionnaire enabled the identification of academic and school mentors’ perspectives and conceptions about ISP and focused on different aspects involved in the process, such as: • The relevance of different internship activities and the degree of collaboration between academic and school mentors in their development. • The responsibility for the definition of the curricular framework of isp. • The aspects included in isp guides. • The relation between the student teacher and the mentors and the kind of activities they are involved in during the whole isp process. • The structure of student teachers’ final report or dissertation; the aspects focused on mentors’ observation and supervision during isp. • The structure and content of the Portfolio of in-school placement teaching practice. Due to the diversity of approaches in the countries involved in the study, the design of the questionnaire took into account the complexity of the ISP process and the multiplicity of aspects involved in the process by offering the respondents rather long lists of the items they should express their position about. As far as the relevance of the activities involved in ISP, all the items were valued above 3,10 in a scale that varied from 1 (nothing) to 5 (very much), regardless of the group (country), what may be seen as the recognition of the diversity and complexity of ISP. Despite the existing significant statistical differences, it is possible to say that there are some items, whose relevance is recognized (generally rated above 4), namely those related to the definition of standards and procedures and those concerning classroom observation, feedback, student-teacher evaluation, and collaborative work involving students and mentors. Collaboration between academic and school mentors throughout the ISP process and regarding the different activities involved is highly valued although the comparison of data regarding the degree of real cooperation and the data regarding the degree of cooperation that should exist suggests the necessity of its enhancement. Regardless of the country, the definition of the curricular framework of ISP depends mainly on the HEI, either the HEI coordinator or their mentors. In-school placement guides tend to include several items, but mentors responding to the questionnaire highlight those related to the definition of the different roles involved and to the teaching practices and their assessment. The planning of activities integrated into the school activity emerges as the most important aspect of the relationship between the student teacher and the mentors. Before the placement, activities concerning class teaching planning and the selection of materials and other resources seem to be the most frequent activities involving mentors, either from HEI or schools and student teachers. During the in-school placement, the most frequent activities involving mentors and student teachers concern not only teaching planning and materials selection but also teaching activities; pupils’ assessment is also highlighted as a frequent activity that involves school mentors and student teachers. Activities developed after the placement are valued in the questionnaire; the most highlighted regard pupils´ assessment and the production of students´ progress reports. The model for the student teacher’s final report or dissertation includes not only the monitoring procedures during its preparation and presentation but also writing rules and procedures, as well as guidelines and rules for the use of data collected by the trainee, the extent/ length of the report and formatting standards and references. The structure and content of this report dissertation are defined at the HEI level
